Characterization of the neuroanatomical variations observed in neurological disorders using a global statistical analysis of brain anatomy.

 

The overall goal of this project is threefold: (1) To define the rate of progress of mild cognitive impairment and Alzheimer's disease; (2) To develop improved methods for clinical trials in this area; and (3) To provide a large database which will improve the design of treatment trials. This project provides information and methods that facilitate effective treatments for Alzheimer’s disease and lead to effective prevention.

We study the covariance of the anatomical structures in the entire brain volume, without segmentation or a priori regions of interest identification. We also provide a global statistical analysis of brain anatomy and identify the relevant shape of deformation patterns that correspond to variations in clinical neuropsychological measures. Our aim is to model the inherent relation between anatomical shape and clinical measures, and to evaluate its statistical significance. The statistical methodology extracts pertinent directions in the momenta space and the clinical response space, in terms of latent variables. We report the results of our analysis on subjects from the Mild Cognitive Impairment group in the Alzheimer's Disease Neuroimaging Initiative (ADNI)
